Recombinant Protein Expression and Purification Assessment Activities
The assessment for the module on Recombinant Protein Expression and Purification will consist of a Construct Design, Expression and Purification Strategy Proposal. Construct Design, Expression and Purification Strategy Proposal For this assessment activity you will choose a protein or protein complex that you plan to express and purify. If you wish to discuss your choice with me, please feel free to come by my office and do so. Write a proposal detailing your strategy to express and purify the protein. In your proposal you should include a description of the protein including some detail of the known biology. Include the amino acid sequence (one letter code) which you should obtain from a protein sequence database. Carry out a bioinformatics investigation of the protein in order to gain some information about the domain structure, sequence homology, possible disordered regions of the protein etc. Using this information design a suitable construct for recombinant expression and purification. Choose a suitable vector and expression host, providing a rationale for your selections. Construct a plan to express the protein with an affinity tag of your choice based on possible tags that I have outlined in the lectures. Design a purification protocol for the protein. Use flow charts and figures to accompany your design design strategy and your purification proposal. Your proposal should be no more than 8 pages in length, including figures and references
